Research Progress on the PI3K/Akt Signaling Pathway and Autophagy Dysfunction in the Pathogenesis of Alzheimer's Disease

Abstract:Alzheimer's disease(AD)is the most common neurodegenerative disease in the world.Its pathogenesis involves multiple pathological processes such as β-amyloid(Aβ)deposition,Tau protein hyperphosphorylation,mitochondrial dysfunction and neuroinflammation.Recent studies have demonstrated that the PI3K/Akt signaling pathway and autophagy dysfunction play a key role in the occurrence and development of AD.This paper systematically reviews the molecular mechanisms by which the PI3K/Akt signaling pathway improves AD symptoms through regulating Aβ deposition and Tau phosphorylation,and inhibiting neuroinflammation.Additionally,this paper discusses the pathological effects of autophagy dysfunction leading to accumulation of damaged mitochondria,increased Aβ toxicity and neuronal death,and it integrates traditional Chinese medicine regulation of the PI3K/Akt pathway and non-drug therapies such as exercise intervention,providing new ideas for the early diagnosis and targeted treatment of AD.
