β1,4-Galactosyltransferase-1 Alleviates Inflammatory Activation of Microglia in Hippocampus by Regulating CD200/CD200R1 Pathway

Abstract:Objective To explore the mechanism of β1,4-galactosyltransferase 1(β4GalT1)on the inflammation and activation of microglia in hippocampus based on the CD200/CD200R1 pathway.Methods Mice were divided into the blank group(n=10,received 0.9% sodium chloride injection),model group(n=10)and inhibition group(n=10).After establishing a mouse microglia inflammation model by peritoneally injecting lipopolysaccharide(LPS),mice in the inhibition group were given targeted injection of the lentivirus LV-si β4GalT1 into the hippocampus.Enzyme-linked immunosorbent assay(ELISA)was used to determine the levels of IL-6,IL-12 and TNF-α in microglia of hippocampus.The mRNA levels of iNOS and CD16,markers of M1 polarization,were detected by quantitative reverse-transcription polymerase chain reaction(qRT-PCR).Hematoxylin and eosin(H&E)staining was used to observe the pathological state of mice hippocampus.Western blotting verified the protein levels of β4GalT1,RCA-1 and CD200R in hippocampal microglia.Results The levels of IL-6,IL-12 and TNF-α in the model group were significantly higher than those in the blank group(P<0.05),which were significantly lower in the inhibition group than the model group(P<0.05).The mRNA levels of iNOS and CD 16 were significantly higher in the model group than he blank group(P<0.05),which were significantly lower in the inhibition group than the model group(P<0.05).H&E staining Results showed that compared with the blank group,mice the model group had deep nucleation,cytoplasmic shrinkage and apoptosis.Compared with the model group,the vertebral cell structure of neurons in the inhibition group was relatively intact,and cell apoptosis was reduced.The protein levels of β4GalT1,lectin RCA-1 and CD200R in the model group were significantly higher than those of the blank group(P<0.05),which were significantly lower in the inhibition group than the model group(P<0.05).Conclusion β4GalT1 promotes the activation of microglia in hippocampus by regulating the CD200/CD200R1 pathway through glycosylation of CD200R1.
