Effects of SDF-1 Expression on Proliferation,Invasion of Hepatocellular Carcinoma Cells and Tumorigenesis in Nude Mice

Abstract:Objective To investigate the effects of stromal cell-derived factor 1(SDF-1)expression on hepatocellular carcinoma(HCC)cell proliferation,malignant invasion and tumorigenesis in vivo.Methods shRNA targeting SDF-1 was transfected into HepG2 cells,and the cells were randomly divided into the Control group,shRNA-NC group,and SDF-1-shRNA1 group.Clone formation assays were performed to evaluate cell proliferation.Cell invasion was assessed using Transwell assays.Western blot was used to detect the protein expression levels of Ki67,Survivin,Vimentin,E-cadherin,and N-cadherin.A nude mouse transplantation model was established to compare tumor proliferation abilities between the Control and SDF-1-shRNA1 groups,and the number of Ki67-and VEGF-positive cells was determined by immunohistochemistry.Results Compared with the Control group,the SDF-1-shRNA1 group exhibited significantly decreased clone formation rates(P<0.05),Ki67 and Survivin protein levels(P<0.05),invasive cell counts(P<0.05),and Vimentin and N-cadherin protein levels(P<0.05).The E-cadherin protein levels were significantly increased(P<0.05).In vivo,the SDF-1-shRNA1 group showed reduced tumor volume and weight(P<0.05)and decreased numbers of Ki67-and VEGF-positive cells(P<0.05).Conclusion Interference with the SDF-1 gene inhibits HepG2 cell proliferation and invasion.
