Expression and Clinical Significance of miR-105 and GOLIM4 in Breast Cancer

Abstract:Objective To investigate the expression of microRNA-105(miR-105)and Golgi integral membrane protein 4(GOLIM4)in breast cancer patients and their relationship with prognosis.Methods Ninety-six breast cancer patients who underwent surgery in Handan Central Hospital from March 2018 to March 2019 were included.Cancer tissues and adjacent tissues were collected.miR-105 expression was detected by qRT-PCR,and GOLIM4 protein expression was detected by Western blot.Pearson correlation was used to analyze the relationship between miR-105 and GOLIM4 in breast cancer tissues.The relationship between miR-105,GOLIM4,and clinicopathological features was analyzed using t-tests or one-way ANOVA,and their association with prognosis was evaluated.Results Compared with paracancerous tissue,breast cancer tissues showed lower GOLIM4 expression and higher miR-105 expression(P<0.05).miR-105 was negatively correlated with GOLIM4 expression(r=-0.458,P<0.05).Patients with histopathological grade G3,lymph node metastasis,TNM stage Ⅲ+Ⅳ,and Luminal A molecular subtype showed higher miR-105 expression and lower GOLIM4 expression compared to patients with grade G1/G2,no lymph node metastasis,TNM stage Ⅰ+Ⅱ,Luminal B,HER-2(+),or triple-negative molecular subtypes(P<0.05).The survival rate in the miR-105 high-expression group[75.51%(37/49)]was lower than in the low-expression group[89.36%(42/47)](Log rank χ2=4.410,P=0.036).Similarly,the survival rate in the GOLIM4 low-expression group[74.00%(37/50)]was lower than in the high-expression group[91.30%(42/46)](Log rank χ2=4.864,P=0.027).Conclusion Breast cancer patients exhibit high miR-105 expression and low GOLIM4 expression,both of which are associated with prognosis.These markers are valuable for prognostic evaluation in clinical breast cancer patients.
