The clinical value of Twist1 gene methylation in the auxiliary diagnosis of bladder cancer

Abstract:Objective To explore the clinical value of Twist1 gene methylation in the auxiliary diagnosis of bladder cancer.Methods The TCGA database was used to compare the expression level of Twist1 between tumors and corresponding adjacent tissues,and to evaluate the prognostic value of Twist1 expression level.Methylation-specific PCR was used to verify the methylation level of the Twist1 gene in bladder cancer patients and healthy people,and to compare the sensitivity and spec-ificity of the clinical diagnostic gold standard and MSP detection results.Results The expression level of Twist1 in bladder cancer was significantly higher than that in the corresponding adjacent tissues.Age had a significant correlation with the overall survival period of bladder cancer in the high-expression group and low-expression group of Twist1(P<0.001).The methylation expression level of the Twist1 gene detected in the urine of bladder cancer was higher than that of healthy people(P<0.01).Cystoscopy and MSP detection results had good sensitivity and specificity(AUC=0.877).Conclusion The Twist1 gene is highly methylated in bladder cancer.The methylation expression level of the Twist1 gene detected in the urine of bladder cancer is higher than that of healthy people.The non-invasive methylation detection of the Twist1 gene provides an auxiliary diagnostic method for the initial diagnosis of bladder cancer.
