Relationship between High-risk HPV Infection and CHD7,BPTF Expres-sion in Tissues of Patients with Cervical Cancer and Cervical Intra-epithelial Neoplasia

Abstract:Objective To investigate the relationship between high-risk human papillomavirus(HPV)infection and expression of chromatin helicase DNA-binding protein 7(CHD7)and bromine domain transcription factor(BPTF)in patients with cervical cancer and cervical intra-epithelial neoplasia(CIN).Methods A total of 104 cervical cancer tissue specimens obtained during cervical biopsy or operation from May 2018 to May 2020 were selected as cervical cancer group,82 tissue specimens of CIN were selected as CIN group,and and 54 normal cervical tissue specimens were selected as control group.The expression of CHD7,BPTF and high-risk HPV in the three groups and the expression of CHD7 and BPTF in cervical cancer patients with different clinicopathological characteristics were compared to explore the relationship of CHD7 and BPTF expression with high-risk HPV infection and prognosis in patients with cervical cancer.Results The positive expression rates of CHD7,BPTF and high-risk HPV gradually decreased in cervical cancer group,CIN group and control group,and the differences between the two groups were statistically significant(P<0.01).The positive expression rates of CHD7 and BPTF in patients with stage Ⅲ or Ⅳ cervical cancer based on the International Federation of Obstetrics and Gynecology(FIGO),low differentiation,invasion depth≥1/2 of muscle layer,lymph node metastasis and high-risk type HPV-positive cervical cancer were higher than those with stageⅠor Ⅱ by FIGO staging,medium and high differentiation,invasion depth<1/2 of muscle layer,no lymph node metastasis and high-risk type HPV-negative cervical cancer(P<0.05,P<0.01).Spearman correlation analysis showed that the expression of CHD7 and BPTF were positively correlated with high-risk HPV infection in cervical cancer patients(r =0.407,0.469,P<0.001).After 3 years of follow-up,the overall survival rate of 104 cervical cancer patients was 43.27%(45/104).The 3-year survival rate of cervical cancer patients with negative expression of CHD7(76.19%,16/21)was higher than that of cervical cancer patients with positive expression of CHD7(34.94%,29/83)(P<0.01).The3-year survival rate of cervical cancer patients with negative expression of BPTF(77.78%,14/18)was higher than that of cervical cancer patients with positive expression of BPTF(36.05%,31/86)(P<0.01).Conclusion CHD7 and BPTF are highly expressed in patients with cervical cancer,and are associated with high-risk HPV infection and prognosis.
