The impact of interferon-βon morphology and protein expression of astrocytes

Abstract:Objective To investigate the role of interferon-beta ( IFN-β) at different times, impact the astrocytes ( AS) cultured in vitro on cell growth state, cell morphology, cell connections and cell cytoskeleton-elasticity associated with protein expression. Methods To divide the cultured AS in vitro into control and experimental groups, and the experimental groups were divided into six groups according to different content of IFN-β(102 U/mL and 103 U/mL) at different times for 24 h, 48 h, and 72 h. To identify AS and observe the changes in cell morphology by immune of luores-cence, the cell morphology was using by atomicforce microscope, the cell junction by electron mi-croscopy and the cell growth was assayed by MTT and analyse the expression of cytoskeletonelasticity associated with protein, including glial fibrillary acidic protein (GFAP),β-actin, cofilin-1, Profi-lin-1, vimentin, by western blot. Results On the effect of different content IFN-β at different times, there's little significant changes of the shape and the number of AS, but there is a obvious in-crease in the cell content sand the expression cytoskeleton-elasticity associated protein. Conclusion IFN-β can enhance the flexibility and toughness of AS, and make the connections of AS more closely. That is important in the central nervous system ( CNS) to maintain the blood-brain barrier permea-bility and prevent the CNS from demyelination. At the same time, IFN-β doesn't make the number of AS increase, but the expression of GFAP increased, which suggests the proliferation of AS, so that it is may form the glial scar to reduce the effect of IFN-β on MS.
