Expressions of filam in A and protein kinase B in small bowel adenocarcinoma and their significance

Abstract:Objective To explore the expression of filamin A (FLNA) and PKB in small bowel adenocarcinoma and to analyze their clinical significance. Methods Reverse transcription polymerase chain reaction (RT-PCR), Immunohistochemistry (SP) RT-PCR and Western blot method were respectively used to detect the expression of FLNA and protein kinase B ( PKB) in 65 speci-mens of small bowel adenocarcinoma and para carcinoma tissues ( 5 cm distant to the margin of small bowel cancer and no cancer infitration in microscope ) . Results The expression of FLNA mRNA was significantly lower in small bowel adenocarcinoma tissues than that in the corresponding normal tissues, while the expression of PKB mRNA was significantly higher in small bowel adenocarcinoma tissues than that in the corresponding normal tissues ( P<0. 05 ) . The expression of FLNA protein was significantly lower in small bowel adenocarcinoma tissues than that in the corresponding normal tissues, while the expression of PKB protein was significantly higher in small bowel adenocarcinoma tissues than that in the corresponding normal tissues ( P<0. 05 ) . Expression levels of FLNA and PKB were correlated with the TNM staging, lymphnode metastasis, and the depth of tumor invasion, but were not correlated with gender, age and the differentiation degree ( P>0. 05) . Spearman rank correlation coefficient showed that the expression level of FLNA was in positive correlation with the PKB level ( r=-0. 289, P=0. 008) . Conclusion The expression of FLNA and PKB proteins may be correlated with the occurrence, development of small bowel adenocarcinoma. Highly activated phosphatidylcholine 3-kinase ( PI3K)/PKB signaling pathway maybe a potential therapeutic target for treatment.
