Neuroprotective effects of troxerutin and cerebroprotein against neuronal damage induced by cerebral ischemia

Abstract:Objective To investigate the neuroprotective effects of troxerutin and cerebropro-tein against neuronal damage induced by cerebral ischmia injury. Methods Male SD rats were di-vided into 3 groups ( n=10):normal control group ( NC) ,middle cerebral artery occlusion reperfu-sion (MCAO/R) group, MCAO/R+(troxerutin and cerebroprotein, TC) group (MCAO/R+TC). One hour after ischemia and 23 hours after reperfusion, 6 rats in each group were evaluated by neu-ral function test and magnetic resonance imaging ( MRI ) , and then sacrificed for determining the cerebral infarction volume with 2,3,5-triphenyltetrazolium chloride ( TTC) staining;the remaining 4 rats in each group were sacrificed, and the protein levels of Caspase 1, Caspase 3,Caspase 8 in the ischemia penumbra were determined by Western Blotting. Results After MCAO/R, cerebral infarc-tion and neurological deficit were manifested. Troxerutin and cerebroprotein was proved to be effec-tive in improving neurological functions and decreasing infarction on MCAO/R animals. Compared with MCAO/R groups, the expression of Caspase 1, Caspase 3, Caspase 8 in NC groups were de-creased significantly ( P<0. 05 ) . Conclusion Troxerutin and cerebroprotein has protective effect against injury induced by cerebral ischemia reperfusion, which is related to their role in inhibiting the neuronal apoptosis.
