Abstract:Objective To investigate the bacterial pathogen-carrying status of Hyalomma asiaticum in Jiuquan,Gansu. Methods During May 2024,free-living H.asiaticum specimens were collected in Jinta County,Jiuquan.Morphological and molecular biological methods were used to identity tick species. The 16S amplicon sequencing method was then used to analyze the diversity of the sample microbial communities.Tick-borne bacterial pathogens of public health significance such as Rickettsia and Bartonella were selected for specific amplification, and carried out phylogenetic analysis after one-generation sequencing. Results Morphological and molecular biological identification verified that all collected samples were H.asiaticum.Analysis of microbial composition indicated that,at the genus level,Francisella and Rickettsia were the predominant genera.Notably,Rickettsia represented a tick-borne pathogen with public health significance.Phylogenetic analysis showed that the Rickettsia carried by H.asiaticumber of the spotted fever group of Rickettsia and a crucial zoonotic pathogen, belonging to Rickettsia sibirica. Conclusion H.asiaticum in the Jiuquan carrying R.sibirica, belonging to the spotted fever group Rickettsia(SFGR).It is important zoonotic pathogen, mainly causing North Asian ticks-borne spotted fever. This research bridges the gap in studies regarding Rickettsia carriage by ticks in this area, and provides important data for the study of H.asiaticum carrying Rickettsia sibirica in Jiuquan.
