Morphology and DNA barcoding identification of 3 flies intercepted firstly at Zhongshan Port

Abstract:Objective To accurately and rapidly identify the intercepted flies from entry ship at Zhongshan Port.Methods Morphology and DNA barcoding techniques used to identify three intercepted flies,one male from a Romanian inbound container,one female from a Macau entry ship,and one male from a Hong Kong entry ship at Zhongshan Port.Results The morphological identification results of the three flies were Phaonia rufiventris,Passeromyia heterochaeta,and Neomyia indica,respectively.Combined with the BOLD database comparison results of DNA barcode sequences,identity of the intercepted flies were confirmed to be 100%in P.rufiventris,99.85%in P.heterochaeta,and 99.85%in N.indica,respectively.Conclusion These three flies were the first intercepted in domestic ports,among them,Phaonia rufiventris being a species not previously recorded in China.The combined use of morphological and DNA barcoding identification methods would enable accurate identification of invasive flies,and provide technical support for port quarantine and inspection.
