Establishment of nucleic acid mass spectrometry for detection of knockdown resistance gene mutations in Aedes albopictus

Abstract:Objective To establish a method for detecting knockdown resistance(kdr)gene mutations associated with resistance to pyrethroid insecticides by nucleic acid mass spectrometry(SVS).Methods Specific primers and probes were designed to achieve multiple amplification in the same PCR reaction system,and the adult Aedes albopictus mosquitoes collected in Tongzhou District,Beijing in August 2023 were selected.Nucleic acid was extracted from a single sample by grinding,and the base mutations at amino acid sites 1016,1532 and 1534 of voltage-gated sodium channel(VGSC)gene were determined by nucleic acid mass spectrometry,and the accuracy of nucleic acid mass spectrometry for the detection of kdr mutations in Ae.albopictus was verified by gold standard next-generation sequencing.Results Nucleic acid mass spectrometry showed that the mutation sites of 44 Ae.albopictus were consistent with the results of first-generation sequencing.Conclusion Mass spectrometry detection of kdr mutations in Ae.albopictus shows a high coincidence rate compared with the results of first-generation sequencing,which is expected to be a new method for detecting VGSC gene mutations in Ae.albopictus.
