Polymorphism and resistance mutation frequency of insecticide target genes in Anopheles sinensis in Neijiang

Abstract:Objective To understand the resistance status of Anopheles sinensis to commonly used insecticides and the mutation of resistance related genes in Neijiang,Sichuan,so as to provide support for the control of Anopheles sinensis in this region.Methods 86 An.sinensis mosquitoes collected from four counties(cities,districts)in Neijiang in September 2018 and July 2021 were subjected to gene fragment sequencing to detect the genotypes of three target genes(ace,VGSC,rdl)and four resistance related loci(ace-119,VGSC-1014,rdl-296,rdl-327).Individual genotype types and frequencies,as well as resistance allele frequencies,were analyzed,and haplotype identification and sequence alignment analysis were performed on VGSC and rdl genes.Results Except for the low resistance frequency of rdl-327(2.94%),the resistance frequencies of ace-119,VGSC-1014,and rdl-296 were all high,at 44.77%,83.14%,and 89.41%,respectively.Genetic haplotype analysis showed that VGSC-H1,VGSC-H2,and VGSC-H3 had different evolutionary origins from VGSC-H4 and VGSC-H5.Conclusion Based on this,it is speculated that the resistance level of An.sinensis in the city is relatively high.It is recommended that professional institutions strengthen scientific guidance on vector control work,and adopt comprehensive control measures for mosquito control.
