Detection and phylogenetic analysis of Babesia spp.in dogs and ticks parasitizing dogs

Abstract:Objective In order to understand Babesia infections in dogs and ticks parasitizing dogs from a police dog base in Nanchang,Jiangxi,and analyze the phylogeny of Babesia in positive samples.Methods From August to October 2022,dog blood samples and ticks parasitizing dogs were collected.Genus-specific primers targeting 18S rRNA,ITS-1,COX1 and HSP70 genes of Babesia species were used for PCR detection and sequence analysis.Results A total of 52 host dogs were investigated,and 47 dog blood samples and 264 parasitic ticks were collected.All of ticks were identified as Rhipicephalus sanguineus and divided into 110 pools.Two tick specimens were detected for Babesia DNA,with a positive rate of 1.82%,whereas the dog blood sample was detected negative.Sequence similarities and phylogenetic analysis based on the amplification of 18S rRNA,ITS-1,COX1,HSP70 genes of Babesia showed that the 2 strains identified in this study and B.vogeli clustered in the same branch.Conclusion There is B.vogeli infection of ticks parasitizing dogs from a police dog base in Nanchang,which poses a potential risk of infection to surrounding animals,prevention and control measures should be strengthened.
