Common potential genes and mechanisms of familial hypercholesterolemia and Alzheimer's disease

Abstract:Objective To investigate the common pathophysiological mechanisms between familial hypercholesterolemia(FH)and Alzheimer's disease(AD).Methods Expression matrices of FH and AD were downloaded from the Gene Expression Omnibus(GEO)database,specifically GSE13985 and GSE5281.Common differentially expressed genes(DEGs)were identified using the limma package.Subsequent analyses included gene function annotation,protein-protein interaction(PPI)network construction,transcription factor(TF),miRNA prediction,and potential drug screening.Results Re-analysis of GSE13985 and GSE5281 microarray data identified 85 common DEGs,with 51 upregulated and 34 downregulated genes.GO and KEGG analyses revealed that these DEGs were significantly enriched in pathways related to oxidative phosphorylation,aerobic respiration,and ATP metabolism.The PPI network was constructed and visualized using Cytoscape,and four core genes(NDUFA6,COX7B,ATP5F1C,and ATP5MG)were identified using the MCODE plugin.Further analysis indicated that NDUFA6,COX7B,and ATP5F1C shared a common transcription factor of ZFP2.Drug prediction using the Enrichr platform identified 1-methyl-4-phenyl-2,3-dihydropyridinium as the top-scoring molecule.Conclusion FH and AD may share common pathological mechanisms.The identification of these common pathways and core genes provides new insights for further exploration of the pathogenesis and potential therapeutic targets for these two diseases.
