Expression and prognosis of transient receptor potential vanilloid family in glioblastoma

Abstract:Objective To investigate the expression of the transient receptor potential vanilloid(TRPV)family in glioblastoma(GBM),its prognosis and its impact on immune cell infiltration in tumor tissues,so as to provide new directions for GBM diagnosis and treatment.Methods Data from databases such as The Cancer Genome Atlas(TCGA),Genotype-Tissue Expression(GTEx),GeneMANIA,STRING,and the Chinese Glioma Genome Atlas(CGGA)were utilized,and the expression of TRPV family members in GBM,their impact on the prognosis of GBM patients,and their relationships with immune cell infiltration and immune checkpoint genes were analyzed by using R(version 4.4.1).Results The mRNA expression level of TRPV4 was found to be significantly higher in GBM tissues compared to normal tissues,while the expression levels of TRPV3,TRPV5,and TRPV6 mRNA were lower.In TCGA-GBM cohort(n=143),patients with higher TRPV4 expression were associated with shorter overall survival(OS)than patients with lower TRPV4 expression{median OS:11.2[95%CI 7.4~13.0]vs 15.0[95%CI 13.0~19.2]months,P=0.022};the results from the CGGA validation cohort(n=137)also supported this observation,with median OS:9.0[95%CI 7.0~15.1]vs 22.7[95%CI 13.9~39.3]months,P=0.006.The differentially expressed genes associated with TRPV4 were significantly enriched in pathways related to cell activity and immune response.Furthermore,high expression of TRPV4 was associated with the accumulation of various immune cells,and,compared to the low-expression group,the expression of most immune regulatory targets was more pronounced in the TRPV4 high expression group.Conclusions The study reveals the important role of TRPV in GBM.TRPV family member TRPV4 is expected to become a biomarker for immune cell infiltration and prognosis,which may provide a new direction for targeted therapy of GBM.
