Expression of transcription factor SOX2 and its significance in gliomas

Abstract:Objective To investigate the expression status of transcription factor SOX2 and its significance in gliomas.Methods Reverse transcription PCR(RT-PCR)and Western blot were used to detect the expression levels of SOX2 mRNA and protein in glioma cell lines U87,U251 and C6,respectively.Immunohistochemical method was used to detect the expression of SOX2 protein in 80 human brain glioma tissues and 10 normal human brain tissues,and the relationship between its expression level and the malignant degrees and pathological grades of gliomas was analyzed.Results Both SOX2 mRNA and protein were highly expressed in U87,U251 and C6 cells.SOX2 protein was highly expressed in human brain glioma tissues,while it was almost not expressed in normal human brain tissues,and there was a significant difference between them(r=0.503,P<0.01);SOX2 protein expression in high-grade gliomas was significantly stronger than that in low-grade gliomas(r=0.450,P<0.01),and its expression also increased significantly with the increasing of glioma pathological grades(r=0.379,P=0.012).Conclusion SOX2 overexpression is closely related to the pathological characteristics of gliomas and may play an important role in the occurrence and development of gliomas.
