Research progress of N-6-methyladenosine regulating protein in glioma

Abstract:N-6 methyladenosine (m6A) is a common chemical modification on RNA molecules in eukaryotes,which significantly affects various biological processes such as RNA stability,translation,and transcription. Although the proportion of m6A modifications in all adenosine is as small as 0.1%-0.4%,it is one of the most abundant epigenetic modifications in eukaryotic mRNA,playing crucial roles in processes such as stem cell renewal,differentiation,transcription,translation,and degradation,as well as in the expression or inactivation of specific genes. The m6A modification is a dynamic and reversible process,regulated by methyltransferases,demethylases,and reader proteins. Core members of the methyltransferase complex include METTL3,METTL14,and WTAP,while FTO and ALKBH5 are known demethylases. Proteins in the YTH gene family,the m6A reader proteins,can recognize and bind to m6A-modified RNA,thereby regulating RNA metabolism and participating in various physiological activities. Studies indicate that the YTH family of m6A reader proteins plays a key role in the formation and development of gliomas. This article aims to review the relationship between m6A and related proteins with gliomas.
