Effects of transcription factor ZBTB20 on PitNET invasive progression

Abstract:Objective To study the effects of the transcription factor ZBTB20 on the invasive progression of pituitary neuroendocrine tumors(PitNET).Methods Tumor sample slices were stained by using immunohistochemical(IHC)method,and ZBTB20 expression levels were measured in these tissues;ZBTB20 overexpressed/knock-down cell lines were established by transfecting ZBTB20/sh-ZBTB20 lentivirus into GH3 cell line;CCK-8 proliferation assay and wound-healing assay were used to measure the effects of ZBTB20 on GH3 cell proliferation and migration;WB and qPCR were used to measure the expression change of proliferation,migration and invasion related to genes.Results IHC staining results showed that ZBTB20 expression was significantly higher in invasive PitNET samples than that in noninvasive samples;in the study of ZBTB20 overexpressed/Knock-down cell lines,CCK-8 proliferation assay and wound-healing assay suggested that ZBTB20 improved the proliferation ability and migration ability of GH3 cell,which was correlated with upregulation of PCNA,MMP2 and MMP14 expression,as shown by WB and qPCR.Conclusion The transcription factor of ZBTB20 is positively correlated with the invasion and development of pituitary tumors.ZBTB20 enhances the proliferation and invasion ability of pituitary tumor cells by promoting the expression of PCNA and MMP proteins,which is of great significance for understanding the mechanism of invasion and development of pituitary tumors and exploring potential therapeutic targets.
