The role of hypoxia microenvironment in the proliferation of human glioma cells

Abstract:Objective The relationship between hypoxia microenvironment and the proliferation of human glioma cells was investigated and Wnt signaling pathway engaged in regulation of proliferation was discussed.Methods The experimental group (culturing U87 cell line tumor cells in 5% oxygen content conditions) and control group (culturing U87 cell line tumor cells in 21% oxygen content conditions) were designed.The cells were set in corresponding incubator.The cell growth was detected with Cell Counting Kit (CCK-8) assay.Limiting dilution assay was conducted to detect the self-renewed capability and monoclonal ability.PCR and Western Blot were carried out to gauge the mRNA and protein expression of markers of the canonical Wnt/β-catenin pathway.Results Compared with the control group,the proliferation of experimental group was more significant,so did the self-renewed capability and monoclonal ability.The expression of mRNA and protein of β-cantenin and C-myc proto-oncogenes (C-myc) were up-regulated and the expression of mRNA and protein of glycogen synthase kinase-3 (GSK-3β) were down-regulated.Conclusion Hypoxia microenvironment promotes the proliferation of glioma cells by activating the canonical Wnt/β-catenin pathway.
