Effect of CCN3 on expression of glioma cells and tumor invasion: a preliminary study

Abstract:Objective The expression of secretary protein nephroblastoma-overexpressed gene (NOV/CCN3) in A172 and U87 gloom cell lines with different malignancy was studied and then the function of CCN3 in glioma cells and the possible molecular mechanism were discussed.Methods Quantificational real-time polymerase chain reaction (qRT-PCR) and enzyme-linked immunosorbent assay (ELISA) were used to compare the expression of CCN3 between A172 and U87cells and their culture media.Human recombinant protein CCN3 was used to treat A172 cells,then MTS assay was used to determine the proliferation and transwell assay was used to determine the invasion of A172 cells after treatment with CCN3.Results The expression of CCN3 in rnRNA level in A172 cells was 15.63 folds lower than that of U87 cells.CCN3 protein in A172 cell culture media was 48.62 pg/mL compared to 549 pg/mL in U87 cell culture media;U87 cells was more invasive than A172 cells and 10 ng/mL CCN3 promoted A172 invasion with no effect on the proliferation.Conclusion CCN3 expression may be closely associated to the malignancy of glioma and have a relationship with invasion of glioma,but it has no effect on the proliferation.
