Protection of allicin on PC12 cells injury induced by 6-OHDA in vitro

Abstract:Objective The protection of allicin on PC12 cells injury induced by 6-hydroxydopamine (6-OHDA) and the possible underlying mechanism are investigated.Methods The injury model of PC12 cells were established by using 6-OHDA and the protective effects of allicin were assessed by cell viability, lactate dehydrogenase (LDH) release and apoptosis analysis.The intracellular reactive oxygen species (ROS) generation and endogenous antioxidative enzyme activities were measured to determine its anti-oxidative activities.Western blot analysis was used to detect the expression of large-conductance Ca2+-activated K+channels (BK) protein and to investigate its role in the protection of allicin.Results Allicin significantly attenuated 6-OHDA-induced decrease of cell viability, inhibited LDH release and reduced apoptotic cell death in PC12 cells.Allicin significantly attenuated 6-OHDA-induced ROS and malondialdehyde ( MDA ) generation, and preserved endogenous antioxidative enzyme activities.And allicin also significantly increased the expression of BK channels, and the pretreatment with BK antagonist paxilline partially reserved the protective effects of allicin.Conclusion Allicin protects PC12 cells against 6-OHDA-induced cytotoxicity through the activation of BK channels and its anti-oxidative activities.
