Effect of physcion on the expression of Bcl-2 and Bax in rats after brain injury

Abstract:Objective The effect of physcion on expression of bcl-2 and Bax in a rat model of human umbilical cord mesenchymal stem cells (HUC-MSCs) transplantation after traumatic brain injury is investigated.Methods All forty Wistar rats aging 2~3 months and weighting 200~250 g were randomly divided into control group (n=10), traumatic brain injury (TBI) group (n=10), HUC-MSCs transplantation group (n=10), HUC-MSCs transplantation+physcion group (n=10).Brain tissues in all the groups were taken for testing at 7 d, 14 d and 21 d after HUC-MSCs transplantation.Polymerase chain reaction (PCR) was used to detect the expressions of bcl-2 and bax.Results The expression of bcl-2 gene was significantly decreased and that of bax was mildly decreased in TBI rats.The expressions of bcl-2 and bax were more significantly decreased in HUC-MSCs transplantation +physcion group. Conclusion Physcion can protect nerve cells by reducing the expression of apoptotic gene bax.
