Research progress on the mechanism of cellular senescence in cisplatin-induced acute kidney injury and renal fibrosis

Abstract:Cisplatin is a first-line chemotherapeutic agent for the treatment of various cancers,but its clinical application is limited due to its nephrotoxicity.Recent studies have found that cellular senescence plays a key role in the progression of cisplatin-induced acute kidney injury(AKI).Cellular senescence is characterized by irreversible cell cycle arrest,and senescent cells can form a senescence-associated secretory phenotype by secreting a variety of inflammatory and profibrotic factors,which alters the surrounding microenvironment and further drives the progression of AKI to chronic kidney disease.Therefore,targeted clearance of senescent cells may serve as a potential therapeutic target for renal protection during cisplatin treatment,while its specific mechanism remains unclear.This review aims to elaborate on the role of cellular senescence in cisplatin-induced AKI and renal fibrosis,hoping to provide a new theoretical basis for the prevention and treatment of cisplatin-induced toxic and side effects.
