Effect of salidroside on the expression levels of serum proline hydroxylase 2,protein phosphatase 2A,and mitogen activated protein kinase in patients with rheumatoid arthritis

Abstract:Objective To analyze the effect of salidroside on the expression levels of serum proline hydroxylase 2(PHD2),protein phosphatase 2A(PP2A),and mitogen activated protein kinase(MAPK)in patients with rheumatoid arthritis(RA).Methods A total of 96 RA patients admitted to Qinghai Provincial Hospital of Traditional Chinese Medicine from December 2022 to July 2023 were selected.They were randomly divided into control group and observation group according to the random number table,with 48 cases in each group.The control group was treated with conventional western medicine,while the observation group was treated with salidroside in addition to conventional western medicine,all for 3 months.The traditional Chinese medicine syndrome scores,clinical symptoms and signs,laboratory indicators[including erythrocyte sedimentation rates(ESR),C-reactive protein(CRP),rheumatoid factor(RF),anti cyclic citrulline peptide antibody(anti-CCP)],clinical efficacy,Health Assessment Questionnaire(HAQ)and Disease Activity Score 28(DAS28)scores and serum PHD2,PP2A,MAPK levels and gene expressions,before and after treatment,as well as adverse reactions was compared.Results After treatment,the scores of the main and secondary symptoms in both groups were lower than those before treatment,and those in the observation group were lower than those in the control group(all P<0.05).After treatment,the clinical symptoms and signs of the observation group were milder than those of the control group,and the levels of ESR,CRP,RF,and anti-CCP were lower than those of the control group(all P<0.05).The total effective rate of the observation group was higher than that of the control group[95.8%(46/48)vs 80.4%(37/46)](P=0.020).After treatment,the HAQ and DAS28 scores in the observation group were lower than those in the control group[(5.0±1.0)vs(7.2±1.2),(3.1±0.5)vs(4.1±0.7)](t=9.528,7.444,both P<0.001).After treatment,the levels and gene expression of PHD2 and MAPK in the observation group were lower than those in the control group,while the level and gene expression of PP2A were higher than those in the control group(all P<0.05).There was no statistically significant difference in the incidence of adverse reactions between the two groups(P=0.528).Conclusion The treatment of RA with salidroside can enhance clinical efficacy,alleviate patients'symptoms,improve laboratory indicators,and be safe.It is speculated that it is related to the decrease of PHD2 and MAPK expression and the increase of PP2A expression.
