The expression changes and clinical significance of pigment epithelium-derived factor in patients with premature ovarian insufficiency

Abstract:Objective To discuss the expression changes and clinical significance of pigment epithelium-derived factor(PEDF)in patients with premature ovarian insufficiency(POI).Methods Totally 10 POI patients admitted to the Fifth Affiliated Hospital of Xinjiang Medical University from January 2020 to December 2021 were selected as the observation group.In addition,20 women with normal and natural menopausal ovarian function evaluation and menopausal age>40 years old were selected as the normal control group and menopausal control group,respectively.Double antibody sandwich enzyme-linked immunosorbent assay was used to detect the expression level of serum PEDF in three groups;the differences in PEDF expression levels were compared among three groups and the relationship between PEDF and various sex hormone indicators was analyzed.Results The serum PEDF levels in the observation group and menopausal control group were significantly lower than those in the normal control group[0.48(0.16,2.49),1.54(0.49,4.10)μg/L vs 7.32(4.22,9.91)μg/L](all P<0.05).However,there was no statistically significant difference between the observation group and the menopausal control group(P>0.05).The follicle-stimulating hormone(FSH)and luteinizing hormone(LH)levels in the observation group were higher than those in the normal control group,while progesterone and anti-Mullerian hormone(AMH)levels were lower than those in the normal control group(all P<0.05).There was no statistically significant difference in testosterone levels among the three groups(P>0.05).PEDF was negatively correlated with FSH and LH(r=-0.410,P=0.003;r=-0.391,P=0.005),and positively correlated with AMH(r=0.498,P<0.001),but no correlation with estradiol,progesterone,and testosterone(r=0.070,P=0.631;r=0.211,P=0.142;r=-0.066,P=0.651).Conclusions The serum PEDF level in POI patients is lower than those in women with normal ovarian function,but there is no significant difference compared to natural menopausal women.PEDF is correlated with ovarian function related hormones.
