Relevance between fecal and intestinal mucosal flora and tight junction protein ZO-1 in patients with functional gastrointestinal disorder

Abstract:Objective To explore the correlation between fecal and intestinal mucosal flora and tight junction protein ZO-1 in patients with functional gastrointestinal disorder.Methods Fecal and intestinal mucosal specimens were collected from 32 patients with functional gastrointestinal diseases from August 2014 to March 2015 in Shanxi Provincial People's Hospital.Bacteria DNA in faece and intestinal mucosa and ZO-1 mRNA in intestinal mucosa were extracted.DNA expressions of bacteroidetes,firmicutes,lactobacillus,enterococcus,enterobacteria and ZO-1 mRNA expression were determined by real time quantitative polymerase chain reaction.The correlation between fecal and intestinal mucosal flora and ZO-1 was analyzed.Results The shannon index in intestinal mucosal flora was significantly lower than that in fecal flora [(2.3 ± 1.2) vs (2.9 ± 0.5)] (P =0.012).Bacteroidetes (45.7%),firmicutes (42.2%) and proteobacteria (8.6%) were dominant bacteria in faece.Proteobacteria(56.6%),firmicutes (20.2%) and bacteroidetes (12.7%) were dominant bacteria in intestinal mucosa.Pearson analysis showed that intestinal bacteroidetes ratio was positively correlated with ZO-1 expression (r =0.378,P < 0.05);fecal bacteroidetes ratio was negatively correlated with ZO-1 expression (r =-0.420,P < 0.05);intestinal lactobacillus ratio and enterobacteria ratio were positively correlated with ZO-1 expression (r =0.530,0.355;P < 0.05).Conclusions There are differences between flora in intestinal mucosa and faece.The correlation between a certain kind of bacterium and ZO-1 expression in intestinal mucosa is different from that in faece.
