Interaction between cardiovascular drug composition gomisin G and warfarin

Abstract：Objective To analyze the interaction between cardiovascular drug composition gomisin G and warfarin.Methods Action sites of gomisin G and cytochrome P2C9 (CYP2C9) were detected by molecular docking method.Combining structures of gomisin G,warfarin and CYP2C9 were established to prove that gomisin G was a good metabolic substrate for CYP2C9.Results The crystal structure of CYP2C9-binding property of gomisin G was confirmed.Warfarin was separated from CYP2C9 active site and gomisin G was binded to the cavity.Gomisin G was tightly combined with CYP2C9 gene Phe476 and Gln214 through hydrogen bond.The length of chemical bond of gomisin G-CYP2C9 was shorter than that of warfarin-CYP2C9,demonstrating that gomisin G is a better substrate for CYP2C9 than warfarin.Conclusion Gomisin G is a good metabolic substrate for CYP2C9;the combination of gomisin G with CYP2C9 can inhibit the combination of warfarin with CYP2C9.
