Regulating action of Chinese patent medicine with similar effect as selective serotonin reuptake inhibitor on 5-hydroxytryptamine system in rats with myocardial infarction and depression

Abstract:Objective To explore the effect of Chinese patent medicine with similar effect as selective 5-hydroxytryptamine (5-HT) reuptake inhibitor on 5-HT system in rats with myocardial infarction and depression.Methods Totally 30 Sprague Dawley rats were randomly divided into control group,western medicine group and Chinese medicine group,with 10 rats in each group.The control group had 2 ml 0.9% sodium chloride by gavage;the western medicine group had 20 mg/kg sertraline + 2 ml 0.9% sodium chloride;the Chinese medicine group had 40 mg/kg Xinling pills + 2 ml 0.9% sodium chloride.Three groups were treated for 3 weeks to make myocardial infarction with depression models;the molding rates were 80.0% (8/10) in all groups;the rats were executed 3 d after molding.Serum level of 5-HT and levels of 5-HT,5-HT2A receptor(5-HT2AR) and 5-HT transporter(SERT) in platelet were detected.Results Serum levels of 5-HT in western medicine group and Chinese medicine group were significantly higher than the serum level of 5-HT in control group [(373 ± 52),(442 ± 23)ng/L vs (182 ± 10)ng/L] (P < 0.05);the serum level of 5-HT in Chinese medicine group was significantly higher than that in western medicine group(P < 0.05).Levels of 5-HT and 5-HT2AR in platelet in western medicine group and Chinese medicine group were significantly higher and the level of SERT in platelet was significantly lower than those in control group [(221 ± 22),(265 ± 29) ng/L vs (181 ± 11) ng/L;(231 ±13),(281 ±49)ng/L vs (206±15)ng/L;(180 ±5),(180±7)ng/L vs (282 ±22)ng/L] (P<0.05);levels of 5-HT and 5-HT2AR in platelet in Chinese medicine group were significantly higher than those in western medicine group (P < 0.05).Conclusion Chinese medicine Xinling pills and western medicine sertraline have similar action in treating myocardial infarction complicated with depression in rats therapeutic.
