Correlation between length of telomere and injury of neural function in cerebral ischemia-reperfusion mice

Abstract:Objective To explore the the correlation between length of telomere and injury of neural function in cerebral ischemia-reperfusion mice.Methods Fifty-six male C57BL/6 mice were randomly divided into middle cerebral artery occlusion(MCAO) group(24 mice divided into 3 groups equally),sham group(24 mice divided into 3 group equally) and blank control group(8 mice).The MCAO group had MACO models by sutureoccluded method(1 h) and reperfusion;the sham group was ligated external carotid artery and common carotid artery for 1 h;the blank control group had no special treatment.At different time points after reperfusion(24,48,72 h),neural function was evaluated by modified Neurological Severity Score (mNSS);genomic DNA was extracted from peripheral blood to detect length of telomere by real-time fluorescence quantitative polymerase chain reaction method.Results The mNSS in MCAO group 24,48,72 h after reperfusion were significantly higher than those in sham group(P <0.05).Length of telomere 72 h after reperfusion in MCAO group was significantly shorter than that in sham group and blank control group [(0.72 ± 0.24) vs (0.97 ± 0.12),(1.04 ± 0.27)] (P < 0.05).Length of telomere was negatively correlated with mNSS 72 h after reperfusion in MCAO group (r =-0.713,P <0.05).Conclusion The length of telomere can be significantly shortened 72 h after MCAO reperfusion in mice;which is associated with severity of neural function injury.
