Effect of isoproterenol on delayed rectifier potassium channel of rabbit superior cervical ganglion neurons in vitro

Abstract:Objective To investigate the effect of isoproterenol(ISO) on electrophysiological characteristics of delayed rectified potassium channel (Ik) in superior cervical ganglion (SCG) neurons of rabbits.Methods Totally 8 specific-pathogen-free male white rabbits were involved;single SCG neurons were acutely isolated.The whole-cell patch clamp technique was used to record changes of Ik in normal SCG neurons (control group,n =10),SCG neurons with 10μmol/L ISO induction 20-60 min(ISO intervention group,n =10)and SCG neurons with 10μmol/L ISO induction 60 min and elution 30 min(ISO elution group,n =10).The Ik current curve,I-V curve and steady-state activation curve were analyzed.Result The amplitude of Ik current curve in ISO intervention group was larger than that in ISO elution group and control group.Peak current densities of Ik in control group,ISO intervention group and ISO elution group were (81 ± 23),(201 ± 81) and (105 ± 18)pA/pF,respectively;there was a significant difference between ISO intervention group and control group (P < 0.05).Half-activation voltages in control group,ISO intervention group and ISO elution group were (9.86 ± 0.40),(3.61 ±0.28) and (6.56 ±0.26) mV;slope factors were (19.66 ±0.53),(15.63 ±0.33) and (15.12± 0.31),respectively;there were no significant differences among groups (P > 0.05).Conclusion ISO has a reversible effect on electrophysiological characteristics of Ik of SCG neurons,it can increase peak current density and change shape of current activation curve.
