Expressions of peripheral HOXA transcript at the distal tip and microRNA-125b in patients with ST-elevation acute myocardial infarction undergoing percutaneous coronary intervention

Abstract：Objective To analyze the expressions of peripheral long-chain non-coding RNA-HOXA transcript at the distal tip (HOTTIP) and microRNA-125b (miRNA-125b) in patients with ST-elevation acute myocardial infarction (STEAMI) undergoing percutaneous coronary intervention (PCI).Methods From June 2015 to February 2016,41 patients with PCI for STEAMI(observation group) and 30 cases without STEAMI (control group) in General Hospital of Ningxia Medical University were enrolled;the observation group was divided into 0-6 h group(11 cases),＞ 6-12 h group(13 cases) and ＞ 12-24 h group(17 cases) according to the duration between STEAMI attack and PCI.Expressions of peripheral HOTTIP and miRNA-125b were detected through real-time fluorescence quantitative polymerase chain reaction.Correlations among HOTTIP,miRNA-125b,cardiac troponin T(cTnT) and high sensitive C reactive protein (hs-CRP) were analyzed.Results Expression levels of HOTTIP and miRNA-125b showed no significant differences among 0-6 h group,＞ 12-24 h group and control group(P ＞ 0.05);the levels in ＞ 6-12 h group were significantly higher than those in control group[(1.09 ± 0.06) vs (0.18 ±0.05),(1.348 ±0.247) vs (0.113 ±0.024)] (P ＜0.05).Expression levels of HOTTIP and miRNA-125b in STEAMI group were significantly higher than those in control group before PCI(P ＜ 0.05);the levels were significantly decreased after PCI [(0.29 ± 0.08) vs (0.78 ± 0.09),(0.093 ± 0.047) vs (0.807 ± 0.043)] (P ＜ 0.05) and there were no significant differences between groups (P ＞ 0.05).After PCI,HOTTIP and miRNA-125b were correlated with cTnT(r =-0.32,P =0.03;r =-0.27,P =0.04);hs-CRP showed no correlations with HOTHP and miRNA-125b(r =-0.13,P =0.31;r =-0.10,P =0.42).Conclusion Expressions of HOTTIP and miRNA-125b increase in STEAMI patients and decrease after PCI;HOTTIP and miRNA-125b may be involved in the process of myocardial ischemia-reperfusion injury in STEAMI.
