Effect of helicobacter pylori on apoptosis and proliferation of different types of gastric cancer cells and normal gastric epithelial cells

Abstract:Objective To explore the effect of helicobacter pylori (Hp) on the morphology,cell cycle,cell apoptosis and proliferation of gastric cancer cells and normal gastric epithelial cells.Methods Inverted microscope was used to observe the morphological changes of human poorly differentiated gastric adenocarcinoma cell line MGC-803,human gastric epithelial cell GES-1,human gastric cancer cell line BGC-823 and human gastric adenocarcinoma cell line SGC-7901 in 2 observation groups (respectively added Hp live bacteria suspension with multiplicity of infection 200/1 and 100/1) and control group (without adding Hp live bacteria suspension) after 24 h and 48 h cultivation.The expressions of apoptosis protein Caspase-9,Caspase-3 and proliferation protein E2F transcription factor-l,P21,P53 were tested by Western blot assay after 24 h infection with 100/1 HP.The proliferation inhibitory rates of 4 cell lines were tested by methyl thiazolyl tetrazolium assay.The cell apoptosis index,proliferation index and cell cycles were tested by flow cytometry.Results After infection with Hp,the normal morphology of 4 cell lines were significantly changed with the increase of time and bacteria numbers.The results of Western blot assay showed that expressions of apoptosis and proliferation proteins of 4 cell lines were changed in different degrees after 24 h infection with Hp.After 48 h infection with Hp,all cell lines were subject to proliferation inhibition,and the proliferation inhibition rates under the influence of 200/1 HP were significantly higher than those under the influence of 100/1 HP [MGC-803 cell:(40.2 ±3.8)% vs (32.7 ±3.2)%,GES-1 cell:(63.7±3.6)% vs (41.2 ±3.5)%,BGC-823 cell:(31.0 ±2.7)% vs (22.7 ±2.2)%,SGC-7901 cell:(32.0 ± 2.9) % vs (27.3 ± 2.5) %] (P < 0.05).After 24 h infection with 100/1 Hp,the apoptosis indexes of 4 cell lines were higher than those in control group [MGC-803 cell:(2.6 ± 0.9) vs (0.5 ± 0.5),GES-1 cell:(4.9±0.8) vs (1.9±0.6),BGC-823 cell:(2.7±1.1) vs (0.7±0.4),SGC-7901 cell:(2.2±0.6) vs (0.9 ±0.6)];the proliferation index of SGC-7901 cell was lower than that in control group [(46.8 ±2.7) vs (52.8 ± 3.6)];the proliferation indexes of MGC-803,GES-1 and BGC-823 cells were higher than those in control group [(43.2±1.7) vs (42.0±1.6),(44.7 ±4.7) vs (32.3 ±2.5),(52.8±2.8) vs (41.6±1.3)],the proportions of G0/G1 phase of these 3 cell lines were lower than those in control group [(57.8 ± 3.6) % vs (65.6±2.8)%,(55.1 ±2.4)% vs (67.5 ±3.6)%,(47.0±3.4)% vs (58.2±4.4)%],the proportions of S phase of these 3 cells were higher than those in control group [(41.3 ± 3.4) % vs (33.2 ± 2.5) %,(29.8 ± 2.4) % vs (19.9 ± 1.6) %,(45.1 ± 3.4) % vs (22.4 ± 2.3) %],the differences were all statistically significant (P < 0.05).ConClusion Hp can induce apoptosis and proliferation disorder of gastric cancer cells and normal gastric epithelial cells.
