Effect of activity of adenosine monophosphate-activated protein kinase on survival of microglial cells after oxygen glucose deprivation and reperfusion injury

Abstract:Objective To study the effect of activity of adenosine monophosphate-activated protein kinase (AMPK) on survival of microglial cells after oxygen glucose deprivation and reperfusion (OGD/R) injury.Methods The microglia BV2 cells were divided into OGD/R group and control group;the OGD/R model was induced by oxygen deprivation (6 h) and reperfusion (24 h).After synchronization of cell cycle in serum free medium for 12 h,the cells were treated with 5,100,50 μmol of 5-aminoimidazole-4-carboxamide (AICAR) to activate the AMPK activation,and 0.1,1,l0 μmol of compound C to inhibit the AMPK activation,respectively.Then four methyl tetrazolium method was used to detect the survival rate of BV2 cells and flow cytometry was used to detect the apoptosis rate.Results The AICAR and Compound C had no significant effect on BV2 cell in control group (P > 0.05);the cell survival rate in OGD/R group was significantly lower than that in control group [(70.8 ± 13.6) % vs (100.0 ± 0.0) %] (P < 0.01);5,50,100 μmol of AICAR increased the cell survival rate [(84.8±5.4)%,(83.3 ± 1.3)%,(82.8 ±2.6)%],while 10 μmol of Compound C reduce the cell survival rate [(28.7 ± 5.3) %] in OGD/R group.The apoptosis rate of cells in OGD/R group was significantly higher than that in control group [(17.3 ± 1.9) % vs (2.7 ± 0.5) %] (P < 0.01),which was significantly increased by 10 μmol of Compound C further [(47.6 ± 4.4) %] (P < 0.01),Conclusion Inhibition of AMPK activity can aggravate the injury of BV2 cells after OGD/R.
