Differentiation of regulatory T cells induced by immature monocyte-derived dendritic cells in peripheral blood of patients with systemic lupus erythematosus in vitro
Abstract:Objective To observe the differentiation of regulatory T cells (Treg) to CD4+ CD25+ forkhead box P3 (FOXP3) + Treg cells induced by immature monocyte-derived dendritic cells (MDDC) in peripheral blood of patients with systemic lupus erythematosus (SLE) in vitro.Methods The immature MDDC in peripheral blood from healthy people(control groups) and SLE patients (SLE groups) were separated and cultured in vitro.The immature MDDC and autologous CD4+ T cells were cocuhured, and the proportion of FOXP3+ CD4+ CD25+ Treg cells were measured by flow cytometry.Results The proportion of FOXP3+ CD4+ CD25+ Treg cells in CD4+ T cells was significantly lower in SLE groups than that in control group [(2.83 ± 0.30) % vs (5.38 ± 0.63) %] (P < 0.05);after being cocultured with immature MDDC, the proportion of FOXP3+ CD4+ CD25+ Treg cells was significantly increased in control group [(8.12 ± 0.70) %] (P < 0.05), but not in SLE group [(3.28 ± 0.27) %] (P > 0.05).Conclusion The MDDC in patients with SLE shows a defective capability to induce the differentiation of CD4+ Treg cells to CD4+ CD25+ FOXP3+ Treg cells.
