Expression and significance of interferon-γ induced protein 10 in serum of patients with acute exacerbation of chronic obstructive pulmonary disease

Abstract:Objective To explore the expression and significance of interferon-γ induced protein 10 (IP-10) in serum of patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD).Methods Fifty-eight patients with AECOPD (observation group) and 28 healthy subjects (control group) from July 2012 to October 2013 were enrolled.The total mRNA in peripheral blood was extracted by TRIzol.Then RNA was reversely transcribed into cDNA by using reverse transcription Kit, subsequently, the mRNA level of IP-10 was detected by real-time polymerase chain reaction (PCR).The expression level of IP-10 protein in serum was detected by enzyme linked immunosorbent assay method.The relation between IP-10 and pulmonary function was analyzed by Pearson's correlation analysis.Results The RNA and protein level of IP-10 in observation group were significantly higher than those in control group [(12.3 ± 4.3) vs (3.8 ± 1.9), (177 ± 35) ng/L vs (87 ± 31) ng/L] (P <0.05).The IP-10 level in serum was negatively correlated with forced expiratory volume in one second (FEV1), forced vital capacity (FVC) and ratio of FEV1 and FVC in percentage (FEV1/FVC) (r =-0.543,-0.486,-0.471, P < 0.05).Conclusion The expression level of IP-10 in serum is increased in patients with AECOPD;the IP-10 level in serum is negatively correlated with pulmonary function and can be involved in the pathogenesis of AECOPD.
