Brain protection effect of gangliosides combined with well points bloodletting in traumatic brain injury model in rats

Abstract:Objective To explore the brain protection effect of gangliosides (GM1) combined with well points bloodletting (BL) in the traumatic brain injury model (TBI) in rats.Methods Seventy five adult SpragueDawley rats were randomly divided into Sham,TBI,GM1,BL,and GM1 + BL group (each n =15).After TBI was induced by electronic controlled cortical impactor,GM1 group was treated with intraperitoneal injection of gangliosides (10 mg/kg,1 time/d),BL group underwent pricking blood of twelve Jing points (2 times/d),GM1 + BL group received above two treatments.The magnetic resonance imaging (MRI) was perform to observe the encephaledema,the modified neurological severity scores (mNSS) were used to assess the behaviors of the animal,the dry/wet weigh method was used to measure the brain water content,and the Evans blue dye (EB) was used to analyze the change of blood brain barrier.Results TBI model in the rats was established successfully.Brain MRI showed that the symptom of brain edema in GM1,BL and GM + BL group was significantly lighter compared with TBI group,especially in GM + BL group.The mNSS,brain water content and EB content were all significantly lower in GM1,BL and GM + BL group than those in TBI group,especially in GM + BL group [(6.0 ±2.0),(6.1 ± 1.5),(2.6 ± 1.1) scores vs (13.3 ±0.7) scores;(78.7 ±0.7)%,(78.8 ±0.8)%,(78.5 ±0.9)% vs(79.9±1.2)%;(215 ±28),(208 ±29),(166±14) μg/g vs (277 ±25) μg/g] (P<0.05).Conclusion Gangliosides combined with bloodletting can effectively improve the prognosis of TBI with good brain protection effect in rats.
