Experimental study on the reversal effect of bortezomib on human acute lymphocytic leukemia cell lines

Abstract:Objective To investigate the possible mechanisms how bortezomib inhibits the proliferation of human acute lymphocytic leukemia cell lines.Methods The Nalm-6 cells were devided into control group cultured with RPMI 1640 medium and durg group cultured with RPMI 1640 medium plus different final density of bortezomib(100,200,400,800,1 600 nmol/L).Different density and different action time of bortezomib were measured to investigate the impact on the growth suppression of Nalm-6 cell.The impact of 400 nmol/L bortezomib on the cell cycle was measured by flow cytometry assay.Results ①After Nalm-6 cell in vitro culture with 100,200,400,800,1 600 nmol/L bortezomib for 12 h,the rate of Nalm-6 cell growth suppression was(8.0 ± 6.1)%,(18.3±7.5)%,(30.3±7.5)%,(37.8 ±2.6)%,(41.1 ±2.1)% respectively;(13.5±5.2)%,(43.3± 6.5)%,(68.7±2.1)%,(81.6±1.6)%,(87.4 ±1.4)% after24 h; (5.21 ±3.71)%,(9.33 ±2.53)%,(49.32±7.12)%,(97.71±1.13)%,(99.93±0.11)% after48 h;(4.00±3.62)%,(4.32±5.61)%,(8.32 ± 4.42) %,(79.52 ± 8.61) %,(99.94 ± 0.13) % after 72 h.The different rate had statistical significance between experimental group and control group(P < 0.05).②After 400 nmol/L bortezomib,cell cycle G2/M phase ratio was(18.6 ± 2.4)% and it was (3.8 ± 2.1)% in the control group; the difference had statistical significance (P < 0.05).Conclusions Bortezomib can inhibit the proliferation of Nalm-6 cell.Bortezomib can block the cell cycle of Nalm-6 cell at the stage of G2/M.
