Protection of puerar on oxidative stress in acute lung injury caused by oleic acid in rats

Abstract:Objective To investigate the protection of puerarin on oxidative stress in acute lung injury induced by oleic acid in rats.Methods Twenty-four Sprague-Dawley male rats were randomly divided into 3 groups:control group,oleic acid (OA) group and puerarin group.Control group was given sodium chloride injection (0.1 ml/kg) by intravenous injection.OA group was given OA (0.1 ml/kg) by intravenous injection.Puerarin group was pre-treated with puerarin(30 mg/kg) intraperitoneally for 30 min before oleic acid infusion.Rat models of acute lung injury induced by oleic acid were used.Morphological changes of lung tissues were observed under light microscope.The content of tumor necrosis factor α(TNF-α) in serum,the activity of superoxide dismutase(SOD) and the content of malonicdialdehyde (MDA) in lung tissues were detected.Results Morphological changes of lung tissues showed that pulmonary inflammation was relieved in puerarin group compared with OA group.Compared with control group,the content of TNF-α increased in serum,the activity of SOD decreased and the content of MDA increased progressively in lung tissues in OA group [(41.7 ± 3.4) ng/L vs (5.8 ± 0.8) ng/L; (78 ± 6) U/mg vs (113 ± 9) U/mg; (11.89 ± 0.64) μmol/g vs (2.49 ± 0.28) μmol/g] (all P < 0.05).Compared with OA group,the content of TNF-α decreased in serum,the activity of SOD increased and the content of MDA decreased progressively in lung tissues in puerarin group [(19.0 ± 1.3) ng/L,(93 ± 7) U/mg,(7.87 ± 0.81) μmol/g] (all P <0.05).Conclusion Puerarin has protective effects on acute lung injury induced by OA,which is probably through enhancing antioxidant activity of organism and decreasing the content of TNF-α.
