Expression of serine/threonine protein kinase B and relationship with inflammation cell factors in skeletal muscle of diabetic rats

Abstract:Objective To evaluate the changes of serine/threonine protein kinase B(PKB),tumor necrosis factor-α(TNF-α)and interleukin 6(IL-6)expression in quadriceps femoris muscle of diabetic rats,and to explore the relationship among them.Methods Adult Wistar rats were divided into experimental group(n =8)and control group(n =8).Diabetic rats in experimental group were fed by high lipoid and glucose diet.After 8 weeks,they were made by a single injection of streptozotocin(STZ)at a dose of 30 mg/kg.After 13 weeks,they were killed for detecting PKB,TNF-α and IL-6 by quantitative immunohistochemistry assay. Correlation analysis was used to evaluate the relationship among them.Results The expression of TNF-α in the control group in skeletal muscle was almost 0 ; the positive cell rates in the experimental group was(3.8 ± 1.4)% and there was a significant difference between two groups(P < 0.05).The expression of IL-6 in normal rat skeletal muscle was(2.0 ±0.8)% ; the positive cell rate in the experimental group was(5.8 ± 0.7)%,which showed a significant difference compared with the control group(P < 0.05).The positive rate of PKB in skeletal muscle of control rats was(9.0 ±0.9)%; the positive cell rate in the experimental group was(2.8 ± 0.8)%,which significantly decreased compared with the control group(P < 0.05). There was a significant correlation among PKB and TNF-α,IL-6(r =-0.776,r =-0.698,P < 0.05).Conclusion PKB pathway can inhibit the expression of the TNF-αand IL-6 in quadriceps femoris muscle of diabetic rats.
