Growth-inhibiting effects of toremifene combined with cisplatin on p53-deficient strain of human lung cancer H1299 cells

Abstract:Objective To study the effects of toremifene combined with cisplatin on human lung cancer H1299 cells(p53-negative).To evaluate the expression of p-glycoprotein(P-gp)in the H1299 cells.Methods The inhibition rate of H1299 cells was evaluated by the 3-(4,5-dimethylthiazo1-2-y1)-2,5-diphenyltetrazolium bromide (MTT)assay.Distribution of cell cycle and rate of apoptosis and the expression of P-gp in the cells were determined by flow cytometry.Results Toremifene(≥20 μmol/L)could inhibit the growth of H1299 cells directly.The inhibitory rate(IR)of the low dose toremffene(5,10 μmol/L)combined with cisplatin were higher than that of cisplatin alone,which was dose-dependent.Toremifene and its combination with cisplatin could increase H1299 cells apoptosis markedly,which was also dose-dependent.Toremifene combined with cisplatin could cause H1299 cells cycle to change.The positive rates of P-gp in H1299 cells were 44.9%.Conclusions The low dose toremifene combined with cisplatin show significant synergistic anti-tumor effects on H1299 cells.Toremifene combined with cisplatin can induce the H1299 cells apoptosis markedly.Its mechanisms can be related with change of phase in cell cycles and apoptosis,which does not depend on p53 gene and estrogen receptor.
