Determination of sunitinib in human plasma by reverse phase high-performance liquid chromatography

Abstract:Objective To develop a reverse phase high performance liquid chromatography (RP-HPLC)method for the determination of sunitinib in human plasma.Methods The plasma samples were extracted with ethyl acetate and separated on an XDB-C18 column,using methanol-0.02mol/L Sodium Dihydrogen Phosphate(70∶30)as the mobile phase.The flow rate was 1.0 mL/min.The fractions were simultaneously detected by diode array detctor with wave length at 391 nm for sunitinib and at 300 nm for internal standard.Results The method was linear for sunitinib in the range of0.5-12mg/L.The correlation coefficient was 0.996 and the lower limit of quantification was 0.5mg/L.The matrix effect was from 93.36% to 104.26%.The extraction recovery was within the range of 73.12% -86.50% (n = 5 ).Conclusion The RP-HPLC method is simple,sensitive and accurate for the determination of sunitinib in human plasma.
