Effect of fructose-1,6-bisphosphate on the myocardial ultrastructure and oxidative stress in Diabetic cardiomyopathy rats

Abstract:Objective To study the effect of Fructose-1,6-Bisphosphate ( FBP) on the myocardial ultrastructure and oxidative stress in rats with STZ-induced diabetic cardiomyopathy(DCM). Methods Diabetes mellitus model in healthy male Sprague-Dawley(SD) rats was induced by single injection of streptozotocin (STZ,Sigma)into intraperitoneal with a dose of 65mg/kg. Intervening subjects( FBP group) were given 500 mg/( kg · d) body weight of FBP intraperitoneal administration, while diabetes mellitus subjects ( DM group) and normal control subjects ( NC group) received 500 mg/( kg · d) of saline After one month, all rats were killed and the myocardial ultrastructures were studied by transmission electron microscopy (TEM). The contents of malondialdehyde(MDA) , activity of superoxide dismutase( SOD), nitric oxide synthase (NOS) and inductive nitric oxide synthase (iNOS) were measured in myocardial homogenate. Results Compared with NC group, more ruptured myofibril was found in DM group, as well as increased mitochondria, vacuolar degeneration and increased electron concentration of capillary basement membrane. At the same time, the concentration of MDA and activation of NOS and iNOS increased (P <0.05) , while the activation of NOS and iNOS in FBP group decreased (P < 0. 05). Conclusion FBP can protect diabetic myocardium and decrease oxidative stress.
