Experimental study on effect of salvia militiorrhiza on microcirculation of indtestines and inflammatory factor in rats with severe acute pancreatitis

Abstract:Objective To explore the effect of salvia miltiorrhiza on intestinal icrocirculatory blood flow and the level of plasma endotoxin and serum concentrations of TNF-α, IL-1β, IL-6 in rats with severe acute pancreati tis(SAP). Methods One hundred and ninety-two male SD rats were randomly divided into control group, SAP group, and treatment group. The rat model of SAP was established by intravenous injection of 4% sodium tauro cholate into the pancreaticobiliary duct. The rats in treatment group were injected with salvia miltiorrhiza via the cer vical vein. Radioactive biomicrosphere technique was performed to measure the intestinal blood flow after 2,6,12,and 24 h,and measured the level of plasma endotoxin and serum concentrations of TNF-α, IL-1β, IL-6, and the pathological changes in intestinal mucosa were observed. Results The intestinal blood flow of SAP group was(0.52 ±0.14) ml/(min · g) ,(0.43 ±0.04) ml/(min · g), (0.38 ±0.53) ml/(min · g), (0.33 ± 0.47) ml/(min ·g) ,respectively] significantly decreased than that of control group[ ( 1.24 ± 0.13 ) ml/( min · g), ( 1.27 ± 0.27 )ml/( min · g ), ( 1.36 ± 0.42 ) ml/( min · g), ( 1. 14 ± 0.63 ) ml/( mi n · g), respectively ] ( P < 0.01 ) at each time point; the level of plasma endotoxin and serum concentrations of TNF-α,IL-1β,IL-6 significantly increased at each time point( P <0.01 ); and the intestinal mucosal injury was more severe( P <0.01 ). The intestines blood flow in treatment groupp[ (0.68 ±0.35)ml/(min · g), (0.64 ±0.28) ml/(min · g) ,(0.33 ±0.47) ml/(min · g) ,re spectively] was higher than that of SAP group at 2,6,and 12 hours( P <0.01 ) ;the level of plasma endotoxin and serum concentrations of TNF-o, IL-1β, IL-6 significantly decreased at 2,6, and 12 hours (P < 0.01 ), but the in crease was not significant at 24 hours; and the pathological changes in intestinal mucosa was improved(P < 0.01 ).Conclusions In SAP rats, the intestinal blood flow decreases and the level of plasma endotoxin, serum concentra tions of TNF-α,IL-1β,IL-6 increases. Salvia miltiorrhiza could improve intestinal microcirculation and alleviate in testinal injuries in SAP rats.
