Expression of PKC-α and Caspase-3 protein and their relation with apoptosis in colorectal carcinomas

Abstract:Objective To study the expression of protein kinase C (PKC)-α and Caspase-3 in colorectal carcinoma and to analyze their relation with apoptosis.Methods Immunohistochemistry and the TdT-mediated dUTP nick and labeling(TUNEL) were used to investigate apoptosis and the alteration of PKC-α and Caspase-3 in 47 cases of colorectal carcinomas,21 colorectal adenomas and 15 normal tissues.Results The positive rates of PKC-α expression in normal colorectal tissues,adenomas and carcinomas were 20.0%,47.6%,66.0%,respectively(P<0.05).Overexpression of Caspase-3 was 31.9 %,61.9%,80.0% in carcinomas,adenomas and normal colorectal tissues,respectively (P<0.05),which was correlated with turnout cellular differentiation (P<0.05).The apoptosis index (AI) of was significantly lower than that of PKC-α-negative cases (1.62±0.57 vs 3.28±0.75) (P<0.05).The apoptosis index of Caspase-3-positive cases was significantly higher than that of Caspase-3-negative cases (2.40±0.55 vs 1.25±0.59) (P<0.05).The expression of PKC-α was negatively correlated with Caspase-3 (r=-0.498,P<0.05).Conclusions The expression of PKC-α and Caspase-3 is closely associated with apoptosis in colorectal carcinoma.The PKC-α and Caspase-3 may participate in the mechanism of apoptosis and play important roles in malignance of colorectal carcinoma.
