Effects of Qian Long Tong Capsule on the expression of TGF-β1 and mRNA in rats of hyperplastic prostatic tissues

Abstract：Objective To observe the effect of Qian Long Tong Capsule on the positive expression of TGF-β1 and TGF-β1 mRNA in hyperplastic prostatic tissues of rats by using immunohistochemistry and in situ hybridization,and to investigate the related molecular mechanisms of Qian Long Tong Capsule in treating benign prostatic hyperplasia at the levels of translation and transcription.Methods A total of 60 male SD rats were selected.10 were randomly selected as the normal control group and other 50 were used to establish the model.After models were successfully established,rats were then randomly divided into model group,Long Bi Shu group and Qian Long Tong Capsule high,middle and low dose groups.Immunohistochemistry and in aitu hybridization were employed to observe the difference in mean gray scale,integral optical density of TGF-β_1 mRNA total area of TGF-β_1-positive cells in all groups.Results The mean gray scale of TGF-β_1 of Qian Long Tong Capsule high,middle dose group and Long Bi Shu group was (2.00±0.69)×10~4,(2.84±0.54)×10~4 and (2.62±0.62)×10~4 respectively,all showing statistical significance compared with that of model group of (3.66±0.89)×10~4 (P＜0.01).The integral optical density of TGF-β_1 of Qian Long Tong Capsule high,middle dose group and Long Bi Shu group was (1.05±0.29)×10~3,(1.74±0.51)×10~3 and (1.76±0.47)×10~3 respectively,all showing statistical significance compared with that of model group of (2.83±1.15)×10~3 (P＜0.01).The total area of TGF-β_1-positive cells of Qian Long Tong Capsule high,middle dose group and Long Bi Shu group was (4.78±0.95)×10~3,(5.76±0.69)×10~3 and (5.63±0.88)×10~3 respectively,all showing statistical significance compared with that of model group of (7.13±0.98)×10~3 (P＜0.01).The integral optical density of TGF-β_1 and total area of TGF-β_1-positive cells of Qian Long Tong Capsule low dose group were (2.14±0.44)×10~3 and (6.36±1.06)×10~3 respectively,beth showing statistical significance compared with those of model group (P＜0.05).The mean gray scale,integral optical density of TGF-β_1 and total area of TGF-β_1-positive cells of Qian Long Tong Capsule high dose group were lower than those of Long Bi Shu group (P＜0.05).Conclusion Qian Long Tong Capsule can decrease the positive expressions of TGF-β_1 and TGF-β_1 mRNA in hyperplastic prostatic tissues.
