Effect of Astragalus Polysaccharides on apoptosis of hematopotietic cells isolated from cord blood

Abstract:Objective To investigate the effect of Astragalns Polysaccharides (APSs) on apoptosis of He-matopotietic cells isolated from cord blood. Methods Mononuclear cells (MNC) were separated from umbilibal cord blood by using Lymphoprep. The cells were cultured in IMDM serum-free medium which was added to APSs of different concentrations. After culturing 48 h and 72 h, we detected the apoptosis rate by AnnexinV-FITC test and the expression of Caspase-3 protein on the harvested cells by using flow cytometry. Results After the cells were cultured in IMDM serum-free medium for 48 h or 72 h, we found that AnnexinV and Caspase-3 protein were all expressed in UBC Hematopotietic cells. After incubation with APSs for 48 h, the apoptosis rate of AnnexinV-FITC and the expression of Caspase-3 in the APSs 40 μg/ml group were lower than those in the control group(P<0.05). After 72 h, the expression of Caspase-3 was decreased greatly at the concentration of APSs 40μg/ml. Conclusions APSs 40μg,/ml can inhibit apoptosis in UBC Hematopotielic cells cultured in IMDM serum-free medium without any GSF. The Caspase-3 is involved in apoptosis of UBC Hematopotietic cells during incubation with APSs in vito.
