Expression of endoplasmic reticulum stress-related proteins in adriamycin-induced acute myocardial injury and its significance
Abstract:Objective To study the expression of ERS-related proteins,GRP78 and CHOP,in adriamycin (ADR)-induced myocardial injury and its significance.Methods Forty C57BL/6J mice were divided into control group (n =20) and ADR group (n =20).The animals in ADR group were injected with ADR (20 mg/kg) to induce acute myocardial injury model and those in control group received saline.LVEF and LVFS were detected by echocardiography.The body weight (BW)and heart weight (HW) were recorded and myocardial tissue samples were stained with HE.Expressions of GRP78,CHOP,Bax and Bcl-2 mRNA and protein were detected by RT-PCR and Western blotting.Results The BW,HW,LVEF and LVFS were significantly lower in ADR group than in control group (P<0.01).Histopathological analysis showed irregular arrangement of myocardial cells,cytoplasm vacuolization,myocardial fibrillar distortion and fragmentation in ADR group.The expression level of GRP78,CHOP and Bax mRNA and protein was significantly higher while that of Bcl-2 mRNA,protein was significantly lower and the Bax/Bcl-2 ratio was significantly higher in ADR group than in control group (P<0.05).Conclusion ERS is involved in ADR-induced myocardial injury by regulating the apoptosis of myocardial cells.
