Expression of PTX3 in smooth muscle cells induced by oxidized low density lipoprotein
Abstract:Objective To study the relationship between PTX3 and atherosclerosis,and the role of PTX3 in proliferation and migration of smooth muscle cells induced by oxidized low density lipoprotein(ox-LDL).Methods Twenty male apolipoprotein E knockout mice were randomly divided into high-fat diet group (n=10) and normal diet group (n=10).The animals underwent aortic ultrasonography after 16 weeks of feeding and were then killed to take aortic tissue samples which were stained with HE and PTX3 immunohistochemistry for detecting the proliferation and migration of smooth muscle cells induced by ox-LDL.The smooth muscle cells of aorta,coincubated with ox-LDL at the concentrations of 0 mg/L,20 mg/L,50 mg/L,100 mg/L and incubated with ox-LDL at the concentration of 100 mg/L after they were subclutured,served as an experimental group with normal smooth muscle cells of atorta served as a control group.The proliferation and migration of smooth muscle cells and PTX3 expression in smooth muscle cells were detected.Results The incidence of atherosclerotic plaques was significantly higher in high-fat diet group than in normal diet group (P<0.01).The proliferation and migration rate of smooth muscle cells and the PTX3 expression level in smooth muscle cells were significantly higher in experimental group than in control group (P<0.05,P<0.01).Conclusion PTX3 is involved in the formation of atherosclerosis and is associated with lipid metabolism.
