Relationship between NADPH oxidase p22phox C242T gene polymorphism and lipoprotein oxidation
Abstract:Objective To study the relationship between NADPH oxidase p22phox C242T gene polymorphism and dyslipoproteinemia in Shanghai Han people.Methods Three hundred and thirty-two Han dyslipoproteinemia patients served as a dyslipoproteinemia group and 305 subjects undergoing physical examination served a control group.The frequency of gene types and allelic gene at the site of C242T gene polymorphism was detected by PCR-RFLP,DNA sequencing,spectrophotometry and ELISA respectively.Relationship between NADPH oxidase p22phox C242T gene polymorphism and dyslipoproteinemia was analyzed.Results The serum levels of TG,TC,LDL,Lp(a),VLDL,ox-LDL and ox-HDL were significantly higher while those of HDL were significantly lower in dyslipoproteinemia group than in control group (P<0.05,P<0.01).The levels of ox-HDL,ox-LDL,MDA and NADPH oxidase activity were significantly higher in subjects carrying CT+TT gene than in those carrying CC gene (521.32±121.56μg/L vs 441.92±134.21 μg/L,P=0.015;14.23±5.34 mg/L vs 10.87±9.05 mg/L,P=0.043;8.75±3.52μmol/L vs 5.28±2.34μmol/L,P=0.003;3.45±2.15 μmol/min·mg vs 1.93±1.13 μmol/ min·mg,P=0.000).The NADPH oxidase p22phox C242T gene polymorphism was not an independent risk factor for dyslipoproteinemia.Conclusion NADPH oxidase p22phox C242T polymorphism is not related with dyslipoproteinemia but with lipid oxidation in Shanghai Han people.
